12-HETE (12-hydroxyeicosatetraenoic acid; primarily 12(S)-HETE) is an arachidonic-acid-derived eicosanoid produced predominantly through ALOX12/12-lipoxygenase, with 12(S)-HpETE as an intermediate. 12-HETE functions as a bioactive lipid mediator and can signal through receptors including GPR31. In cancer, increased ALOX12–12-HETE signalling can promote tumour-cell migration, epithelial–mesenchymal transition, invasion, angiogenesis and metastatic dissemination, including interactions between platelets and circulating tumour cells. The typical cancer-associated direction is therefore generally up, while the desired anticancer modulation is generally down. 12-HETE also participates in inflammatory and vascular signalling and has been reported as increased in some Alzheimer-related lipidomic models, although its specific causal role in Alzheimer’s disease remains less established.
