ALDOB (aldolase B; fructose-bisphosphate aldolase B) is a glycolytic and fructolytic enzyme that cleaves fructose-1-phosphate into dihydroxyacetone phosphate and glyceraldehyde and also catalyzes reversible reactions involving fructose-1,6-bisphosphate. ALDOB is highly expressed in liver, kidney and small intestine and links fructose metabolism with glycolysis, gluconeogenesis and central carbon metabolism. Its role in cancer is context-dependent. In colorectal cancer liver metastases, ALDOB is strongly upregulated and enables metastatic tumour cells to exploit fructose as a carbon source, supporting glycolysis, biosynthesis and metastatic growth; ALDOB knockdown reduces liver metastatic growth in experimental models. Conversely, ALDOB is reduced in some cancers, including gastric cancer, where restoration of ALDOB has suppressed glycolysis, proliferation and migration. Therefore,
