| Ku70 - X-Ray Repair Cross-Complementing Protein 6 Abbreviation: Ku70, XRCC6 Type: DNA double-strand break repair protein / NHEJ component Function: Ku70 forms a heterodimer with Ku80 (XRCC5) that rapidly binds DNA double-strand break ends and initiates classical non-homologous end joining (NHEJ). The Ku70/Ku80 complex protects broken DNA ends and recruits DNA-PKcs and additional repair proteins required for DNA-end processing and ligation. Ku70 also participates in telomere maintenance, apoptosis regulation, and cellular DNA-damage responses. Cancer: ↑ Increased Ku70/XRCC6 expression or NHEJ activity can enhance tumor-cell survival following DNA damage and contribute to resistance to radiotherapy and DNA-damaging chemotherapy. High XRCC6 expression has been associated with tumor progression and poorer prognosis in several cancers. However, loss of Ku70-mediated DNA repair can also increase genomic instability and cancer susceptibility, so its role differs between tumor initiation and treatment of established cancers. |
