Function: OTUB2 removes ubiquitin modifications from selected protein substrates and thereby regulates protein stability, signaling, DNA-damage responses, metabolism, and cell proliferation. OTUB2 can modulate K48- and K63-linked ubiquitination and participate in regulation of DNA double-strand break repair.

Cancer: ↑ Frequently increased or oncogenic in multiple cancers. OTUB2 can stabilize oncogenic substrates including PKM2 and KRT80, promoting glycolysis, PI3K/AKT signaling, proliferation, migration, invasion, and metastasis. OTUB2 inhibition or knockdown suppresses tumor-cell growth and progression in several experimental cancer models.
