Type: Transcription factor / cell-cycle regulator

Function: E2F4 is a member of the E2F transcription-factor family that regulates genes involved in cell-cycle progression, DNA replication, differentiation, and quiescence. E2F4 frequently forms repressive complexes with RB-family proteins including p107/RBL1 and p130/RBL2 and is an important component of the DREAM complex that suppresses cell-cycle gene expression in quiescent cells.

Cancer: ↓ Typically reduced or functionally impaired as a cell-cycle repressor. Loss of E2F4-mediated transcriptional repression can promote inappropriate expression of S-phase and proliferation-associated genes. However, E2F4 can show context-dependent oncogenic activity in some tumors.
