Function: FGFR4 is a transmembrane receptor tyrosine kinase that binds fibroblast growth factors and regulates cell proliferation, survival, differentiation, metabolism, and tissue homeostasis. In the liver, FGFR4 forms an important signaling complex with FGF19 and the co-receptor β-Klotho (KLB), regulating bile-acid synthesis and metabolic homeostasis. Activated FGFR4 can signal through pathways including RAS/RAF/MEK/ERK, PI3K/AKT, JAK/STAT3, and GSK3β/β-catenin.

Cancer: ↑ Frequently overexpressed or aberrantly activated in selected cancers, particularly hepatocellular carcinoma. Activation of the FGF19-FGFR4 signaling axis promotes tumor-cell proliferation, survival, resistance to apoptosis, invasion, and treatment resistance. FGF19 amplification or overexpression can create an FGFR4-dependent tumor phenotype, making FGFR4 a therapeutic target for selective kinase inhibitors.
