Arachidonic Acid Pathway - Arachidonic Acid Metabolism / Eicosanoid Pathway

Function: The arachidonic acid pathway generates a large family of bioactive eicosanoid signaling molecules. Arachidonic acid is released from membrane phospholipids primarily by phospholipase A2 (PLA2) and is subsequently metabolized through three major enzymatic branches: cyclooxygenase (COX), lipoxygenase (LOX), and cytochrome P450 (CYP) pathways. These pathways generate prostaglandins, prostacyclin, thromboxanes, leukotrienes, lipoxins, HETEs, and EETs that regulate inflammation, vascular function, immune responses, proliferation, apoptosis, migration, and tissue homeostasis.

Cancer: ↑ / dysregulated. Increased arachidonic acid release and eicosanoid metabolism are common features of many cancers. Pro-tumor branches including COX-2/PGE2, 5-LOX/leukotriene signaling, 12-HETE, CYP4A/20-HETE, and selected CYP-derived EET pathways can promote proliferation, survival, angiogenesis, immune evasion, invasion, and metastasis. However, individual arachidonic acid metabolites can exert opposing effects, so specific enzymes and metabolites should be recorded separately whenever possible.
