NGB - Neuroglobin

Type: Heme-containing globin / oxygen- and redox-responsive cytoprotective protein

Function: Neuroglobin is a predominantly neuronal heme protein involved in cellular responses to hypoxia, ischemia, oxidative stress, and mitochondrial dysfunction. NGB can modulate reactive oxygen and nitrogen species, mitochondrial signaling, calcium homeostasis, energy metabolism, and apoptotic pathways and generally promotes neuronal survival under metabolic or oxidative stress.

Cancer: ↑ Frequently increased in several cancers and may function as a cytoprotective survival factor. Elevated NGB can help tumor cells tolerate hypoxia, oxidative stress, mitochondrial dysfunction, and apoptotic signaling, thereby supporting cancer-cell survival and potentially treatment resistance.

Alzheimer's Disease: ↓ / protective when increased. Reduced or inadequate NGB-mediated neuroprotection may contribute to vulnerability to oxidative and mitochondrial injury, whereas increased NGB expression protects against amyloid-β toxicity, mitochondrial dysfunction, oxidative stress, and neuronal apoptosis in experimental models. Therapeutic NGB upregulation is therefore generally considered favorable.
