Function: FNDC5 is a membrane-associated protein expressed in skeletal muscle, brain, heart, and other tissues. Proteolytic cleavage of its extracellular domain generates irisin, a secreted signaling peptide associated with exercise-induced metabolic and neuroprotective effects. The PGC-1α/FNDC5/irisin pathway regulates energy metabolism, neuroplasticity, synaptic function, inflammatory signaling, and expression of neurotrophic factors including BDNF.

Cancer: ↕ Context-dependent. FNDC5/irisin signaling can influence tumor-cell metabolism, proliferation, apoptosis, invasion, and the tumor microenvironment, but reported effects vary considerably among cancer types and experimental models.

Alzheimer's Disease: ↓ Reduced or impaired FNDC5/irisin signaling is associated with Alzheimer's disease and cognitive dysfunction, whereas increased FNDC5/irisin signaling is generally neuroprotective. Experimental activation of this pathway improves synaptic plasticity and cognition and can reduce neuroinflammation, oxidative stress, amyloid-β pathology, and tau-related abnormalities.
