Function: MUC5AC is a large, heavily glycosylated secreted mucin that contributes to formation of the protective mucus layer on epithelial surfaces. It regulates epithelial barrier properties, cell adhesion, host-pathogen interactions, inflammatory signaling, and interactions between epithelial cells and the extracellular environment.

Cancer: ↕ Context-dependent, with increased expression in several major adenocarcinomas. MUC5AC is frequently overexpressed or aberrantly expressed in pancreatic, esophageal, ovarian mucinous, gastric, and selected lung cancers. Elevated MUC5AC can promote tumor-cell survival, stemness, migration, invasion, immune interactions, and tumor progression. In pancreatic cancer, MUC5AC can enhance integrin αvβ5/Src/STAT3/KLF4 signaling and maintain cancer stem-like properties. However, expression and prognostic significance vary substantially by tumor type.
