SLIT2 - Slit Guidance Ligand 2

Type: Secreted guidance protein / ROBO receptor ligand

Function: SLIT2 is a secreted extracellular guidance protein that signals primarily through ROBO-family receptors. It regulates cell migration, cytoskeletal organization, axon guidance, vascular development, immune-cell trafficking, epithelial integrity, and tissue morphogenesis. SLIT2/ROBO signaling can influence pathways involving small GTPases, β-catenin, PI3K/AKT, and migration-associated signaling.

Cancer: ↕ Context-dependent, with a frequently tumor-suppressive role. Reduced SLIT2 expression or promoter hypermethylation has been reported in several cancers. Restoration of SLIT2/ROBO signaling can inhibit epithelial-mesenchymal transition, migration, invasion, and metastasis. However, SLIT2 can also support angiogenesis or tumor progression in selected tumor and stromal contexts.
