| SYN3 - Synapsin III Abbreviation: SYN3, Synapsin III Type: Neuronal phosphoprotein / synaptic vesicle-associated protein / neurodevelopmental regulator Function: Synapsin III is a neuron-enriched phosphoprotein involved in neurogenesis, axonogenesis, neurotransmitter release, dopaminergic signaling, and regulation of synaptic vesicle function. Compared with Synapsin I and II, SYN3 has particularly important roles during early neuronal development and is also found at extrasynaptic sites such as neuronal cell bodies and growth cones. Alzheimer's Disease: ↓ SYN3 expression has been reported in hippocampal CA1 pyramidal neurons in mild cognitive impairment and Alzheimer's disease, consistent with impaired presynaptic and synaptic function. Favorable Direction in Alzheimer's Disease: ↑ Restoration or preservation of normal SYN3 expression/function may be favorable where reduced SYN3 reflects synaptic dysfunction. Cancer: ↕ Context-dependent. In glioblastoma, SYN3 has demonstrated tumor-suppressive activity. SYN3 expression is reduced in glioblastoma relative to normal tissue and other glioma subtypes, while experimental SYN3 overexpression reduces glioblastoma stem-cell proliferation and stemness and promotes neuronal-like differentiation. Favorable Direction in Glioblastoma: ↑ SYN3 expression/activity appears favorable based on current experimental evidence. |
