GSTM1 - Glutathione S-Transferase Mu 1

Type: Phase II detoxification enzyme / glutathione transferase / antioxidant and xenobiotic metabolism enzyme

Function: GSTM1 catalyzes conjugation of reduced glutathione to electrophilic compounds, facilitating detoxification and elimination of carcinogens, environmental toxins, therapeutic drugs, and reactive products of oxidative stress.

Cancer: ↕ Context-dependent. Normal GSTM1 activity can protect against carcinogenesis by detoxifying reactive carcinogens and oxidative products, and the GSTM1-null genotype has been associated with increased susceptibility to several cancers. However, in established tumors, GSTM1-mediated detoxification can also protect cancer cells from oxidative stress and anticancer drugs and may contribute to treatment resistance.

Favorable Direction in Cancer: Context-dependent. ↑ GSTM1 activity may be protective during cancer prevention by increasing carcinogen detoxification, whereas ↓ GSTM1 activity may enhance sensitivity to certain chemotherapies in established tumors.

Genetic Relevance: A common homozygous deletion produces the GSTM1-null genotype, resulting in absence of functional GSTM1 enzyme activity. This polymorphism can modify susceptibility to environmental carcinogens and alter drug response.

Interpretation Note: Distinguish GSTM1 expression/activity from GSTM1 genotype. “GSTM1-null” means the gene is deleted and enzyme activity is absent; it is not simply reduced expression.
