| NFKBIZ - Nuclear Factor Kappa B Inhibitor Zeta Abbreviation: NFKBIZ, IκBζ, I-kappa-B-zeta Type: Atypical nuclear IκB-family protein / NF-κB transcriptional regulator / inflammatory signaling regulator Function: NFKBIZ encodes IκBζ, an inducible nuclear regulator of NF-κB-dependent transcription. It interacts particularly with NF-κB p50 and p52 homodimers and can either activate or repress selected inflammatory and immune-response genes. NFKBIZ is strongly induced downstream of Toll-like receptor and IL-1 signaling and contributes to regulation of cytokines, antimicrobial genes, and immune-cell differentiation. Cancer: ↕ Context-dependent. ↑ NFKBIZ is oncogenic in several lymphoid malignancies, particularly activated B-cell-like diffuse large B-cell lymphoma (ABC-DLBCL), where increased IκBζ supports NF-κB-dependent survival and inflammatory gene expression. In solid tumors, NFKBIZ can have either tumor-promoting or tumor-suppressive effects depending on tissue and signaling context. Favorable Direction in Cancer: Context-dependent. ↓ NFKBIZ may be favorable in tumors such as ABC-DLBCL that depend on IκBζ-driven NF-κB signaling, whereas loss of NFKBIZ may be unfavorable in cancers where it exerts tumor-suppressive effects. Inflammation: ↑ NFKBIZ generally promotes expression of a subset of NF-κB-dependent inflammatory genes downstream of TLR and IL-1 signaling, although it can also repress selected targets. Interpretation Note: NFKBIZ/IκBζ should not be treated as equivalent to classical IκBα/NFKBIA. IκBζ is predominantly a nuclear transcriptional regulator and can enhance rather than simply inhibit NF-κB signaling. |
