| TPH1 - Tryptophan Hydroxylase 1 Abbreviation: TPH1, TPH-1 Type: Enzyme / serotonin biosynthesis enzyme / aromatic amino acid hydroxylase Function: TPH1 catalyzes the rate-limiting step in peripheral serotonin (5-HT) synthesis by converting tryptophan to 5-hydroxytryptophan (5-HTP), which is subsequently converted to serotonin. TPH1 is expressed primarily in peripheral tissues, including enterochromaffin and other serotonin-producing cells. Inflammatory Bowel Disease: ↑ TPH1 activity and peripheral serotonin production can promote intestinal inflammation. Increased 5-HT signaling has been associated with greater severity of experimental colitis. Favorable Direction in Intestinal Inflammation: ↓ TPH1 expression or activity can reduce peripheral 5-HT production and intestinal inflammatory signaling. Luteolin: ↓ TPH1 gene and protein expression, accompanied by ↓ MAPK/ERK signaling and ↓ 5-HT production in stimulated cells; luteolin also reduced elevated colorectal 5-HT in experimental colitis. Pathway Relationship: Tryptophan → TPH1 → 5-HTP → 5-HT. TPH1 ↓ → serotonin synthesis ↓ → intestinal inflammatory signaling ↓. Interpretation Note: TPH1 should be distinguished from TPH2. TPH1 primarily regulates peripheral serotonin synthesis, whereas TPH2 is predominantly responsible for neuronal serotonin synthesis in the central nervous system. |
