Function: Synaptotagmin-1 is a major calcium sensor on synaptic vesicles that triggers rapid calcium-dependent vesicle fusion and neurotransmitter release through interactions with the SNARE machinery.

Alzheimer's Disease: ↓ Synaptic SYT1 expression or function is associated with impaired presynaptic vesicle release and synaptic dysfunction. Altered SYT1 levels have also been investigated in cerebrospinal fluid and blood as biomarkers of synaptic degeneration in Alzheimer's disease and mild cognitive impairment.

Favorable Direction in Alzheimer's Disease: ↑ Preservation or restoration of normal synaptic SYT1 expression/function is generally favorable.

Biomarker Note: Increased extracellular or CSF SYT1 may reflect synaptic injury and should not be interpreted the same way as increased functional SYT1 within intact presynaptic terminals.
