Carboxylesterase 1 (CES1; hCE1) — Major hepatic serine hydrolase involved in the hydrolysis and metabolism of ester-containing xenobiotics, drugs, prodrugs, and endogenous lipids. CES1 can influence drug activation/inactivation and lipid metabolism. In hepatocellular carcinoma, CES1 has been implicated in a CES1–PPARα/γ–SCD signaling axis associated with tumor progression and chemoresistance; CES1 inhibition can impair mitochondrial function and increase sensitivity to cisplatin.

Typical modulation in cancer: ↑ in some cancers, particularly hepatocellular carcinoma, where elevated CES1 activity may support lipid metabolism, tumor survival, and chemoresistance. However, effects are cancer- and drug-dependent because CES1 can also activate or deactivate ester-containing therapeutics.
