| CLDN4 (Claudin-4) — Integral membrane tight-junction protein involved in epithelial barrier function, cell polarity, and regulation of paracellular transport. CLDN4 can also participate in intracellular signaling outside of classical tight junctions and is a high-affinity receptor for Clostridium perfringens enterotoxin (CPE). Typical modulation in cancer: ↑ CLDN4 is common in many epithelial malignancies and may promote tumor growth, proliferation, EMT, stemness, invasion, and resistance to drug penetration. However, reduced CLDN4 can also accompany loss of epithelial differentiation and EMT in some contexts, so direction should be interpreted by tumor type and experimental setting.
