| DHCR7 (7-Dehydrocholesterol Reductase) — Endoplasmic-reticulum membrane oxidoreductase that catalyzes the reduction of 7-dehydrocholesterol (7-DHC) to cholesterol, a terminal step in cholesterol biosynthesis. DHCR7 regulates cellular sterol composition, cholesterol availability, membrane biology, and downstream lipid-dependent signaling. Typical modulation in cancer: Frequently ↑ in cancer and generally tumor-promoting in several contexts. Elevated DHCR7 has been associated with increased proliferation, invasion, migration, metastasis, altered tumor immunity, and poorer prognosis. In colorectal cancer, DHCR7 ↑ promotes cholesterol accumulation and activates PI3K/AKT/mTOR signaling, whereas DHCR7 inhibition suppresses tumorigenic phenotypes. Typical modulation in Alzheimer’s disease: Context-dependent. DHCR7 influences cholesterol and 7-DHC metabolism, both of which can affect neuronal membrane composition, oxidative stress, and sterol-dependent signaling. However, a consistent AD-specific disease direction is not established. Additional relevance: DHCR7 can also influence ferroptosis through sterol metabolism and susceptibility to lipid peroxidation. |
