| TRAF6 (TNF Receptor-Associated Factor 6) — Signaling adaptor and RING-domain E3 ubiquitin ligase that mediates signaling downstream of Toll-like receptors (TLRs), IL-1 receptors, TNF receptor-superfamily members, and other immune receptors. TRAF6 catalyzes predominantly K63-linked polyubiquitination and promotes activation of TAK1, IKK/NF-κB, MAPK, inflammatory, survival, and stress-response pathways. Typical modulation in cancer: ↑ TRAF6 is frequently tumor-promoting and can enhance NF-κB, MAPK, survival, proliferation, migration, invasion, inflammatory signaling, and treatment resistance. TRAF6 inhibition or downregulation is therefore generally considered anticancer, although effects can vary by tumor type and immune context. Typical modulation in Alzheimer’s disease: ↑ TRAF6-mediated inflammatory signaling may contribute to neuroinflammation through TLR/IL-1/NF-κB pathways. Excessive TRAF6 activity is generally considered potentially detrimental in chronic neuroinflammatory states, although TRAF6 also participates in normal innate-immune and neuronal signaling. |
