RCC1 (Regulator of Chromosome Condensation 1) — Guanine nucleotide exchange factor (GEF) for the small GTPase RAN. RCC1 is bound to chromatin and generates nuclear Ran-GTP, establishing the Ran-GTP gradient that regulates nucleocytoplasmic transport, nuclear-envelope dynamics, mitotic spindle assembly, chromosome condensation, and cell-cycle progression.

Typical modulation in cancer: ↑ RCC1 is frequently associated with increased proliferation, mitotic activity, tumor progression, and poorer prognosis in several cancers. Cancer cells can depend on the RCC1–RAN signaling axis for nuclear transport and mitosis, so RCC1 inhibition or downregulation is generally considered potentially anticancer, although effects remain tumor-context dependent.
