| Mitochondrial Complex V (ATP Synthase; F1F0-ATP Synthase) — Final oxidative-phosphorylation complex of the mitochondrial inner membrane. Complex V uses the proton-motive force generated by respiratory-chain Complexes I, III, and IV to drive phosphorylation of ADP to ATP. It consists of a membrane-embedded F0 proton channel coupled to the catalytic F1 ATP-synthase domain. Typical modulation in cancer: Context-dependent. ↑ Complex V activity can support ATP production and survival in tumors dependent on oxidative phosphorylation, while inhibition of ATP synthase can cause ATP depletion, mitochondrial dysfunction, metabolic stress, ROS changes, and cancer-cell death. Some tumors exhibit altered expression or activity of ATP-synthase subunits as part of metabolic reprogramming. Typical modulation in Alzheimer’s disease: ↓ Complex V activity or expression can contribute to impaired mitochondrial ATP production, energetic failure, synaptic dysfunction, and neuronal vulnerability. ATP-synthase dysfunction and oxidative modification of its subunits have been reported in neurodegenerative disease. Typical mechanistic relationship: Proton gradient / mitochondrial membrane potential → proton flow through F0 → rotation of ATP synthase → ADP + Pi → ATP. |
