HAGH (Hydroxyacylglutathione Hydrolase; Glyoxalase II, GLO2) — Second enzyme of the glyoxalase pathway. HAGH/GLO2 hydrolyzes S-D-lactoylglutathione to D-lactate and regenerates reduced glutathione (GSH), thereby completing detoxification of the reactive glycolytic by-product methylglyoxal. Cytosolic and mitochondrial isoforms are present in human cells.

Typical modulation in cancer: Context-dependent. GLO2 participates in methylglyoxal detoxification and glutathione recycling, but its cancer role is less uniform than GLO1. Increased GLO2 has been associated with proliferation and treatment resistance in some cancer models, whereas mitochondrial GLO2 can also participate in pro-apoptotic signaling, including interaction with BAX. Therefore, no universal cancer direction should be assigned.

Typical modulation in Alzheimer’s disease: Potentially protective through methylglyoxal detoxification, glutathione regeneration, mitochondrial protection, and reduction of dicarbonyl/oxidative stress, but a consistent AD-specific disease direction is not established.

Typical mechanistic relationship: Methylglyoxal + GSH → GLO1 → S-D-lactoylglutathione → GLO2/HAGH → D-lactate + regenerated GSH.
