| Glycine (Gly) — Nonessential amino acid involved in protein synthesis, one-carbon metabolism, nucleotide synthesis, glutathione production, heme synthesis, and mitochondrial metabolism. Glycine also functions as an inhibitory neurotransmitter and as a co-agonist at NMDA receptors. Typical modulation in cancer: Context-dependent. Rapidly proliferating cancer cells may increase glycine uptake and utilization to support purine synthesis, one-carbon metabolism, glutathione production, and biomass generation. Increased glycine metabolism can therefore support proliferation in some tumors, although glycine supplementation may have different effects depending on tumor type and metabolic context. Typical mechanistic relationships: Glycine → serine/glycine one-carbon metabolism → nucleotide synthesis; glycine + glutamate + cysteine → glutathione synthesis; glycine → NMDA receptor co-agonism. b>Recommended group: Core Metabolism/Glycolysis (tgid=4). |
