Pyrimidine / Pyrimidine Metabolism — Metabolic pathway responsible for synthesis, salvage, and degradation of pyrimidine nucleotides including uracil, cytosine, and thymine derivatives. Pyrimidine metabolism supplies UTP, CTP, dCTP, and dTTP required for RNA synthesis, DNA replication, membrane phospholipid synthesis, glycosylation, and cellular proliferation.

Typical modulation in cancer: ↑ pyrimidine synthesis and salvage are commonly associated with rapidly proliferating tumor cells because of increased demand for DNA and RNA precursors. Inhibition of pyrimidine metabolism can suppress proliferation, induce replication stress, cause DNA damage, and enhance sensitivity to anticancer therapies.

Typical mechanistic relationships: De novo pyrimidine synthesis → UMP → UDP/UTP → CTP and dTMP/dTTP → RNA/DNA synthesis. Key pathway enzymes include CAD, DHODH, UMPS, TYMS, and related salvage enzymes.
