| LIMK2 (LIM Domain Kinase 2) — Serine/threonine kinase that regulates actin cytoskeleton organization, cell morphology, migration, invasion, and mitosis. LIMK2 phosphorylates cofilin proteins, thereby inhibiting their actin-depolymerizing activity and promoting stabilization/reorganization of actin filaments. Typical modulation in cancer: Frequently ↑ or hyperactivated and generally associated with increased migration, invasion, cytoskeletal remodeling, metastatic potential, and in some cancers proliferation or therapy resistance. LIMK2 inhibition can reduce motility and invasive behavior, although effects are tumor-context dependent. Typical mechanistic relationship: RhoA / ROCK → LIMK2 activation → cofilin phosphorylation ↑ → cofilin activity ↓ → actin filament stabilization/remodeling ↑ → migration and invasion ↑. Additional functions: LIMK2 also participates in spindle formation, cytokinesis, cell-cycle regulation, and stress-response signaling. |
