Beta-Tubulin (β-Tubulin; TUBB family) — Structural protein family that heterodimerizes with alpha-tubulin to form microtubules. Microtubules are essential components of the cytoskeleton and mitotic spindle and participate in chromosome segregation, intracellular transport, cell morphology, migration, and division.

Typical modulation in cancer: β-Tubulin expression and isotype composition can be altered in cancer. Disruption of β-tubulin polymerization or microtubule dynamics can inhibit mitosis and induce cell-cycle arrest and apoptosis. Increased expression of specific isotypes, particularly TUBB3, is associated in several cancers with aggressive behavior and resistance to microtubule-targeting chemotherapy.

Typical mechanistic relationship: Alpha-tubulin + beta-tubulin → microtubule polymerization → mitotic spindle formation and chromosome segregation. Microtubule destabilization or excessive stabilization → spindle dysfunction → mitotic arrest → apoptosis / cell death.

Drug relevance: β-Tubulin is a major target of taxanes, vinca alkaloids, colchicine-site microtubule inhibitors, and several benzimidazole compounds. Different drug classes either stabilize or destabilize microtubules but can ultimately disrupt mitosis.
