MSH2 (MutS Homolog 2) is a gene that plays a crucial role in the DNA mismatch repair (MMR) system. This system is responsible for correcting errors that occur during DNA replication, such as base-base mismatches and insertion-deletion loops. When the MSH2 gene is mutated or dysfunctional, it can lead to an accumulation of genetic errors, which increases the risk of developing certain types of cancer.
Tumors that exhibit a deficiency in mismatch repair (dMMR), often due to the loss of MSH2 and other MMR proteins, are characterized by a high level of microsatellite instability (MSI).
